Intended use

The Real-time RT-PCR Detection Kit for SARS-CoV-2 is intended to be used to achieve qualitative detection of SARS-CoV-2 viral RNA in various human samplesThe kit is designed to simultaneously detect specific fragments of the ORF1ab and N gene of SARS-CoV-2, and human RNaseP gene (RP), as an internal control of assay quality. This kit is intended for use by trained personnel for auxiliary diagnosis and epidemiological monitoring of SARS-CoV-2.

Testing principle

Samples with viruses can be lysed and viral nucleic acids be departed from proteins, and then be enriched by the beads. Multiple RNase inhibitors help deactivates nucleases to prevent nucleic acids from degradation. Wash and ethanol remove impurities to acquire high purity. After eluted by EB, nucleic acids can be achieved for downstream applications including PCR, NGS and Northern bolt.

This kit designed specific primers and probes for ORF1ab and N genes in the highly conserved region of the novel coronavirus (SARS-CoV-2) genome. Based on the TaqMan fluorescence quantitative PCR platform, the novel coronavirus (SARS-CoV-2) was quantitatively detected by one-step real-time fluorescent RT-PCR technology. The 5 'end of the probe is labeled with FAM and VIC fluorescein respectively. At the same time, internal standard primers and probes are designed based on the conserved region of human RNaseP gene, the 5 'end of the internal standard probe is labeled with CY5 fluoresceinthat can be used to monitor respiratory nucleic acid quality and the PCR system to avoid blank samples and false negative results. With components such as reverse transcriptase and hot-start Taq DNA polymerase, viral RNA is first reverse-transcribed to form cDNA, and then entered into PCR amplification. SARS-CoV-2 ORF1ab and N protein genes were quantitatively detected in nasopharyngeal swabs, throat swabs, alveolar lavage fluid or sputum samples.
